Particle trajectory simulation - spiral microfluidic

Hello! I am currently designing a spiral microchip for CTC isolation from blood sample. I tried doing the particle trajectory simulation but the isolation failed and as I checked the velocity field is not centered. Anyone knows why? Is it because I input the wrong velocity?

The dimension of the chip is as follow: -Height: 110 mikrom -Width: 500 mikrom -Inner circle: 9 cm -Outer radius: 12 cm -Turns: around 1.75

Meanwhile the inlets are seperated in a size of 1:2

I put in a velocity of 2 for the smaller inlet and 3 for the bigger one

Please let me know how to estimate the right velocity in a spiral microfluid.
